ナノセグメント固定化細胞培養基板の調製と幹細胞培養と分化制御

Outline of Annual Research Achievements

幹細胞の未分化状態での保持、培養並びに適切な組織細胞への分化誘導は、幹細胞を用いる次世代の医療技術である再生医療に必須である。幹細胞の運命(未分化状態の保持、特異的組織細胞への分化)の制御は、幹細胞培養基板の物理的特性(剛性 /弾性(stiffness))並びに生物化学的特性(細胞接着部位ペプチドの種類並びに表面密度)に大いに依存することを明らかとした。。様々な剛性 /弾性を有する基板を作成し、その基板上に細胞接着因子並びにペプチド(RGDS, CS-1, DEGA, IKVAV, PDSGR, YIGSR等)を固定化させた基板を調製した。様々なナノセグメント固定化細胞培養基板上において、ES細胞、iPS細胞、造血幹細胞並びに間葉系幹細胞を培養して、(1)未分化状態で培養させる最適な基板の剛性 /弾性並びに最適な細胞接着因子ペプチドあるいは細胞外マトリックス蛋白質(ECM)を判明させた。さらに、(2)適切な組織細胞(骨芽細胞)への分化効率の高い最適な基板の剛性 /弾性並びに最適な細胞接着因子ペプチドとECMを判明させた。造血幹細胞では、フィブロネクチン並びにフィブロネクチン由来CS1ペプチド固定化した弾性率20KPa前後のハイドロゲル基板が最も増殖性に優れていた。間葉系幹細胞を高純度で純化させる膜純化法を開発することができた。ヒトiPS細胞並びにヒトES細胞の培養が20継体以上可能なオリゴビトロネクチン固定化ハイドロゲル基板(弾性率30KPa)を本研究の概念より開発することができた。
