Outline of Final Research Achievements

Treatment with RANKL induces osteoclastogenesis in murine and canine bone marrow cells as well as murine RAW264.7 cells. Analyses of Mitf isoform expression indicated that Mitf-E gene was induced within 24 hours after RANKL treatment in all examined cells. Knockdown of Mitf-E by its siRNA transfection inhibited RANKL-induced osteoclastogenesis. In mouse cells, co-treatment with TGF-β enhanced RANKL-induced osteoclastogenesis, whereas TGF-β inhibited canine osteoclastogenesis induced by RANKL.
