久山町住民における遺伝・環境要因を考慮したインスリン分泌能の長期追跡

Research Abstract

本年度は,久山町の2002年度の生活習慣病健診受診者を対象に、インスリン分泌の指標であるHoMA-βと各SNPsとの関係を横断的に解析した。この健診には40歳以上住民の77. 6%にあたる3,328名が受診した。受診者の96%にあたる3, 196名から同意を得てDNAを抽出し,理化学研究所で遺伝子解析を行った。最終的に,N0TCH2, ADAM30, PKN2, G6PC2-ABCB11, SYN2-PPARG, PPARG, IGF2BP2, ADAMTS9, UBE2E2, FLJ39370, CDKALl, VEGFA, JAZFl, SLC30A8, CDKN2A/2B, CDC123-CAMK1D, IDE/KIFll/HHEX, TCF7L2, EXT2/ALX4, MTNRIB, KCNQl, KCNJll, L0C387761, TSPAN8-LGR5, C2CD4A-C2CD4B, FTOの25遺伝子上の32SNPSを測定した。インスリン分泌の指標であるHoMA-βと各SNPとの関連を検討すると,統計学的に有意な関連を示したものはcDKAL1、IGF2BP2、KCNJll、cDKN2A/2Bの遺伝子上のSNPsであった。

SNPの測定は完了しており、統計解析を行う準備は整っている。

今後、2007年の健診結果からHOMA-βの値を算出し、5年間にわたるインスリン分泌の低下度と各SNPsとの関連を検討する予定である。

Expenditure Plans for the Next FY Research Funding

2007年のインスリン値の測定は完了しているが、電子化作業が遅れたため、予算の執行が遅れた。
今後早期に、2007年度の電子化作業を行って、データセットを整備し、解析を行う予定である。
