Outline of Annual Research Achievements

近年マウス線維芽細胞に２種類の遺伝子を導入するのみで、人工肝細胞を樹立することが可能であると示された。我々は今回申請する研究において、マウス線維芽細胞より人工肝細胞を樹立し、その細胞移植系を利用した新規のC型肝炎ウイルス(HCV)感染モデル動物をマウスにおいて樹立し、その知見を基盤に新規のHCV感染モデル動物の構築を試みる研究を行い、下記の成果を得た。
１．マウス人工肝細胞の樹立、培養、in vitroでの形質解析：マウス肝臓から肝幹･前駆細胞をセルソーターで高度に濃縮し、形質を解析することで、Wnt5aが胆管形成を負に制御すること(Hepatology 2013)、下流分子はCaMKIIでありその活性化がWnt5aにより制御されることを見いだし、報告した(第21回肝細胞研究会で報告)。
２．マウス人工肝細胞を用いた移植モデルの開発：我々は移植した初代幹細胞・初代肝幹･前駆細胞によって高度なドナーキメリズムが得られる細胞移植系を独自に開発し報告した。本移植系での細胞移植効率はMMP活性に依存することを報告した（JDDW2014及びAASLD2014にて発表）。
３．ヒトHCVレセプター強制発現によるHCV感染モデルの構築と人工肝細胞誘導法の検討：人工肝細胞にヒトCD81及びヒトoccludin遺伝子の強制発現を行い、移植によるHCV感染性の評価を試みた。本研究では、十分な感染性の獲得が認められなかった。肝特異的遺伝子の強制発現によって人工肝細胞を誘導しうるか検討を行ったが、肝特異的遺伝子の上昇は技術的に困難で、同様の報告も原法の発表者らが報告したため、さらなる改良を要すると考えられた。
４．ヒト人工肝細胞を用いた移植モデルの開発及びHCV感染モデルの構築に関して検討を行ったが、十分な置換率を得るには至らず、今後の研究課題にて継続して検討する必要があると考えられた。
