酸化的DNA塩基損傷修復と熱ショック蛋白修飾による心血管リモデリング抑制の研究

Outline of Annual Research Achievements

本研究は、酸化的DNA塩基損傷修復(base excision repair:BER)機構の活性修飾と熱ショック蛋白(Heat Shock Protein: HSP)の相加的な心血管保護効果を利用して心血管リモデリングを制御する新規手法の確立をめざすものである。本研究の標的多機能タンパクはApurinic/apyrimidinic Endonuclease 1(APE1)であり、動脈硬化性心血管病変に対する新たな治療法の確立をめざした。
APE1は、核内において酸化的DNA塩基損傷の修復を担い、レドックス制御の転写活性を促進させ、核外において酸化ストレス軽減効果を発揮する。我々は血管リモデリングに必須の内皮前駆細胞(EPCs)を用いて、その機能解析とリモデリングに及ぼす影響を明らかにした。マウス骨髄からEPCsを採取調整し、ヒト末梢白血球ゲノムからクローニングしたAPE1を遺伝子発現用アデノウイルスに組み入れ、これを用いてAPE1過剰発現細胞を調整した。また、APE1特異的SiRNAを作製し、lipofectamin法にて核酸導入してAPE1発現抑制細胞を調整した。
EPCsはAPE1を豊富に発現するが、過酸化水素の暴露でその接着能は濃度依存性に減弱したが、APE1過剰発現細胞では、接着能低下が阻止され、逆にAPE1発現抑制細胞では接着能低下が促進された。APE1のレドックス制御特異的阻害の検討により、これらの機序には局所の活性酸素消去機構の亢進が重要な役割を担うことを明らかにした。
最後に、in vivo実験としてマウスのワイヤー血管障害モデルを用い、障害血管局所にAPE1過剰血管内皮前駆細胞を導入したところ、局所の新生内膜肥厚は、APE1により有意に抑制された。以上より、APE1を介する血管リモデリング抑制が、動脈硬化性心血管病変の新たな治療法となる可能性を明らかにした。
