Outline of Final Research Achievements |
Cell culture-based experiments were done using with esophageal squamous cell carcinoma (ESCC) cells, namely TE4 and TE-8, under 5-aminolevulinic acid in their culture media. By irradiating the ALA-treated ESCC cells with light-emitting diode (LED) TE-8 cells underwent a potent growth inhibition and cell cycle arrest on G1 phase in 24 hours after LED-irradiation, while this inhibitory effect was merely observed in TE-4 cells. Although analysis of cell cycle-related protein expressions by western blot is under investigation at this point, the expression of p53 or p21 did not seem to change in the early phase of post-LED irradiation period. Animal experiments resulted in insufficient examination for the designated three years, however, photodynamic therapy with ALA should be attractive for the treatment of ESCC as well as other photosensitizers such as photofrin that have bibliographically demonstrated their excellent therapeutic effects.
