Ｓｏｘ９を活性化する薬剤を用いた軟骨欠損・変性疾患に対する治療法の開発

Research Abstract

SOX9は軟骨基質の形質維持に極めて重要であり、SOX9の発現を促進することにより軟骨変性の防止あるいは変性軟骨の修復が期待できる。
マウス軟骨前駆細胞株ATDC5にSOX9プロモーター領域を挿入したベクターをトランスフェクションし定常発現株を作成した。ATDC5はインスリン添加により未分化間葉系細胞から軟骨細胞へ分化することが知られている。作成した定常発現株も通常のATDC5と同様にインスリン添加により軟骨多段階分化モデルが作成できることを確認した。
SOX9を活性化する薬剤を同定する目的で1200種類のスクリーニング用薬剤（Prestwick Chemical社）を用いたルシフェラーゼアッセイによる薬効1次スクリーニングを行い候補薬剤を48種類に絞り、2次にて16薬剤、さらに3次にて濃度依存性を認めた4薬剤に絞った。4次スクリーニングとして、正常ATDC5に4薬剤を添加し、定量的RT-PCR 法によりSOX9およびその下流に位置するII型コラーゲンおよびアグリカンのmRNA 発現を検討した。また、軟骨のプロテオグリカンを評価するためアルシアンブルー染色を行い、吸光度測定により定量化した。その結果、抗アレルギー剤Xがコントロール（ジメチルスルホキシドのみ添加）と比較して10～100μMの範囲で濃度依存性にSOX9、II型コラーゲンおよびアグリカンのmRNAを上昇させ、さらにプロテオグリカン含有量を増加させていた。
