Outline of Final Research Achievements |
Several immune system cell surface receptors are reported to be associated with osteoclastogenesis. Dectin 1, a lectin receptor for β-glucan, is found predominantly on cells of the myeloid lineage. In this study we have developed a delivery system for oligonucleotides using schizophyllan (SPG), a polysaccharide that belongs to the β-glucan family. Application of complex consisting SPG and TNF receptor associated factor 6 (TRAF6) specific siRNA to osteoclast progenitor cells inhibited the osteoclast formation in vitro. On the other hand, we also demonstrated that the dectin-1 agonist curdlan potentially inhibits osteoclast differentiation, especially nuclear factor of activated T cell cytoplasmic 1 (NFATc1) expression, and that Syk kinase plays a crucial role in the transcriptional pathways. These data suggest that application of this new complex and curdlan administration could be potential candidates for the treatment of osteoclast-related diseases such as osteoporosis.
