分泌型マイクロＲＮＡは骨格筋可塑性のバイオマーカーとして有効か

Research Abstract

循環血液中に存在する分泌型microRNA（miRNA）が骨格筋の可塑性の指標となるか検討することを目的として、2年計画で実施する。初年度である本年度は、マウスヒラメ筋廃用性萎縮モデルを用いて、ヒラメ筋の萎縮とその後の再荷重による再成長に伴う血漿中のmiRNAをマイクロアレイにて網羅的に解析した。その結果、筋萎縮に伴い2倍以上増加したmiRNAは105種、2分の1未満に低下したmiRNAは38種であった。逆に、対象群には認められず筋萎縮時にのみ認められたmiRNAは75種、その後4週間継続した発現が認められたものは22種であった。対照群に発現が認められるのにもかかわらず筋萎縮時に消失したmiRNAは9種認められた。萎縮後の再荷重2週目にのみ発現が認められたmiRNAは7種、逆に再荷重2週目にのみ消失したmiRNAは37種類であった。また、萎縮直後ならびに再荷重2週目にのみ発現が認められたmiRNAは18種であった。一方、ヒラメ筋におけるmiRNAで、対象群では認められないが、萎縮時にのみ認められるmiRNAは25種、逆に対照群に認められるものの萎縮時には確認できないものは64種、その内で萎縮直後にのみ発現が認められないものは19種、それぞれ認められた。。また、萎縮直後と再荷重2週目にのみ認められるmiRNAは21種、再荷重2週目にのみ消失するmiRNAは5種観察された。こうしたヒラメ筋の量的変化に伴い血漿中と筋肉中で共に特徴的な変化を示すmiRNAがバイオマーカー候補が確認された。今後、miRNA発現量の定量評価ならびにマウス筋芽細胞C2C12を用いた骨格筋可塑性に特異的な分泌型miRNAを特定し、運動に対する骨格筋応答のバイオマーカーの確立を目指すための基礎資料が得られた。
