トキソプラズマにおける宿主オルガネラリクルート因子の同定

| Research Abstract |
トキソプラズマは宿主細胞に侵入した後、宿主細胞機能を様々に修飾することが知られている。中でも特に形態学的に顕著な変化として、宿主のミトコンドリアやERを原虫が増殖している寄生胞近辺に引き寄せる(リクルートする)という現象が昔からよく知られてい る。この現象を引き起こす因子として、原虫が宿主細胞に侵入する際に、原虫の侵入とは独立して宿主細胞に注入される一群のタンパ ク質、ロプトリータンパク質群の関与が強く疑われているが、本年度はロプトリータンパク質を含めた原虫由来のどのタンパク質が宿主オルガネラのリクルートに関わっているのかを解明するために、定量可能な質量分析法であるiTRAQを用いて宿主ミトコンドリアに特異的に結合するトキソプラズマ由来タンパク質の網羅的な同定を試みた。原虫を感染後3時間、12時間、24時間経過した細胞から精製したミトコンドリアと、非感染細胞と原虫の混合試料から精製したミトコンドリアをiTRAQにより比較した。感染細胞ミトコンドリアで非感染細胞の1.5倍以上の存在量を示し、かつ、シグナル配列などのドメイン構造や相同性検索、また各種の既報のプロテオーム解析の結果を参照することで、リクルート因子候補タンパク質として118種類のタンパク質を同定することができた。さらに、ミトコンドリアのリクルートは感染後1～2時間ですでに起きていることが報告されているので、申請者はこのうち感染3時間後にすでに存在量に違いがみられた28種類のタンパク質を最終的なリクルート因子候補タンパク質として同定した。
