Outline of Annual Research Achievements

平成２６年度に二次イオン質量分析計で故障が発生し、修理のために部品を交換したほか、テストを複数回繰り返して実施したことにより、分析が遅れていた。故障個所の特定作業が長期間にわたり、後期の大半で分析不可能な状態に置かれたことから予定していた海底下微生物細胞のＮａｎｏＳＩＭＳ分析が遅れていた。平成２７年度には、その遅れていたＮａｎｏＳＩＭＳ分析を実施した。試料としては南太平洋環流域、および下北半島沖海底下試料を用い、本課題において確立した細胞分離手法を活用して海底下試料に存在する大量の堆積物粒子、すなわち微生物以外の非生物粒子と生物細胞の粗分離を実施した。その後、ＤＮＡ染色剤であるＳＹＢＲ　Ｇｒｅｅｎ Ｉを高濃度で存在させることで得られる非生物粒子と微生物細胞の蛍光色の違いを利用し、ベックマン社製のＭｏｆｌｏ高速セルソーターを用いてＦｌｕｏｒｅｓｃｅｎｃｅ Ａｃｔｉｖａｔｅｄ　Ｃｅｌｌ　Ｓｏｒｔｉｎｇ（ＦＡＣＳ）により微生物細胞のみを試料台上の極小スポット（約１００μｍ程度）に分離・濃縮した。このようにして調製した試料は微生物細胞のみを含むため、高精度なＮａｎｏＳＩＭＳ分析を実施することが可能となり、微生物の代謝活性を検出・定量することが出来た。特に下北半島沖試料については、培養によって活性化した微生物細胞が炭酸、または酢酸を取り込んでいる様子を明確に可視化することに成功し、海底下試料中に存在した微生物の代謝活性を明らかにすることに貢献した。
