| Research Abstract |
1.Lymphoblasoid cellsを用いたGWAS(Stage 1)のデータ収集と解析
HapMapプロジェクト(http://www.hapmap.org/)で採取されたlymphocyteをEB virusで不死化したlymphoblastoid cellsを用い,lymphoblastoid cellを用いて,敗血症を模したcytokine mixture(TNF-alpha, IL-1beta, IFN-ganmma[各2.5ng/mL], CpG[12.5uM])で刺激.刺激6h後のRNAをlymphoblastoid cellより抽出し,IL-6遺伝子発現レベル,細胞培養液中のIL-6濃度を測定する.これら遺伝子発現・蛋白レベルの量的形質に対するGWASを改変して行う.GWAS解析ソフトはPLINKv1.07(http://pngu.mgh.harvard.edu/purcell/plink/)を使用する.
2.重症敗血症日本人コホートのサンプル収集
重症敗血症コホートの患者サンプルおよび臨床データの収集中である.
