好熱性真核生物のゲノム情報を基盤とした小胞体関連分解装置の構造生物学研究

Research Abstract

本研究は、小胞体において糖タンパク質の運命決定を下す小胞体品質管理マシーナリー(フォールディング装置、選別輸送装置、小胞体関連分解装置)の分子認識および作動機構の解明を目的としている。
本研究の特徴の一つは、真核生物の中で高い生育温度(約60℃)を持つ好熱カビのゲノム情報を活用することにある。最終年度では、好熱カビの小胞体関連分解装置に加えて、UGGT、グルコシダーゼII、カルネキシンなどを中心としたフォールディング装置の構成因子のcDNAクローニング、大量発現系の構築、結晶化を行った。その結果、糖タンパク質のプロセッシングに関わるグルコシダーゼIIの触媒サブユニットの結晶構造解析に初めて成功した。これにより、グルコシダーゼIIの基質認識に関する有益な知見を得ることが出来た。
