平成26年度は、薬物/Eudragit S100について,その溶解メカニズムの評価を行った。固体分散体からの薬物及びポリマーそれぞれの溶出速度を同時に評価した。また、固体NMR測定により固体分散体の分子状態を評価し、薬物の溶出挙動に及ぼす影響を検討した。HPLCの検出器にUV及び蒸発光散乱検出器(ELSD)を併用することで、噴霧乾燥により調製されたフェニトイン/S100固体分散体及びグリペングラミド/S100固体分散体からの薬物及びS100の溶出速度を同時に評価可能であった。本手法を用いることにより、固体分散体の溶解時に薬物が非晶質状態を維持する場合は、薬物がポリマーの溶出に伴って溶出し、溶出速度が改善されることを見出した。また、緩和時間測定を含めた固体NMR測定により、固体分散体中における薬物とポリマーの分子レベルの混和性が薬物の溶出性に強く影響することを明らかとした。固体分散体製剤の設計において薬物の溶出性制御は重要な課題であり、本研究で得られた知見は、ポリマー選択や薬物/ポリマー配合比の設定などの処方検討の際に極めて有力な指針となる。今後、他の薬物/ポリマー固体分散体についても同様の検討を行うことで、固体分散体からの薬物溶出メカニズムの更なる解明が期待される。
