翻訳後修飾アセチル化に着目した「新規関節リウマチ検査」の確立

Research Abstract

関節リウマチ診断のための臨床血液検査では、血清中の抗シトルリン化ペプチド抗体・リウマチ因子の検出などが採用されているが、十分とは言えない。従って、従来の検査項目を補完するような新規検査項目が必要とされている。申請者は、前年度の研究で、関節リウマチ患者の末梢血においてalpha-enolase (ENO1)とイソクエン酸デヒドロゲナーゼのアセチル化が亢進していることを突き止めた。そこで、平成25年度は、1)ENO1のアセチル化の測定法の確立を含め、2)診断的意義を明らかにすることを目的とした。
上記1)の目的を達成するにあたり、アセチル化ENO1の診断指標としての有用性を評価する目的で、アセチル化ENO1の「アセチル化を受けているリジン残基とその周辺のアミノ酸配列」を認識する抗体を作製するために、関節リウマチ特異的に亢進しているENO1のアセチル化部位（リジン残基）の特定を試みた。被験試料であるENO1の調製法（ENO1量やプロテアーゼの種類など）や質量分析の条件等を複数検討し、関節リウマチ由来末梢血リンパ球由来ENO1のアセチル化部位の特定を試みたが、アセチル化ペプチドの検出は困難であった。次に、上記2)を遂行するにあたり、ENO1が、関節リウマチ特異的な自己抗体の抗原となりうるかについて調べるために、ENO1およびアセチル化ENO1を抗原とし（アセチル化ENO1は、リコンビナントで作製したENO1に、in vitroでアセチル化酵素およびアセチルCoAを作用させ、効率的にアセチル化させることで調製した。）、関節リウマチ患者血清を用いて、ウエスタンブロッティングを行ったところ、ENO1に反応する血清がいくつか認められが、アセチル化依存的な反応強度の変化は認められなかった。
