卵巣明細胞腺癌におけるメタボローム解析とオートファジーの役割

Research Abstract

「卵巣明細胞腺癌では高発現しているHNF1βがアミノ酸トランスポーターSLC3A1を介して代謝を制御し、酸化ストレスに関与している」ことを示すために以下の研究を行った。
まず、HNF1βの発現抑制細胞株をメタボローム解析した。この結果により明細胞腺癌ではHNF1βを介して嫌気的糖代謝が促進されていることがわかった。また、アミノ酸トランスポーターSLC3A1の輸送物質であるシスチン濃度や抗酸化ストレス物質であるグルタチオンがHNF1β発現抑制株では低いことがわかった。培養液中のシステイン（シスチンの前駆物質）濃度依存性に細胞内のグルタチオン濃度が変化することを確認した。SLC3A1発現抑制株では細胞内のグルタチオン濃度が低くなり、活性酸素種が多くなることを確認した。
以上の実験は卵巣明細胞腺癌細胞株であるRMG2細胞株とJHOC-5細胞株とで確認した。
