天然由来抗酸化色素によるぶどう膜炎軽症化の検討

Outline of Annual Research Achievements

研究実施計画に則り、ラットにエンドトキシン誘導ぶどう膜炎(EIU)を惹起し、ウニ殻由来抗酸化色素エキノクロームを投与してそ の抗炎症効果を検討した。その結果、エキノクロームの投与時期、投与量、投与経路を変えて検討し、安全かつ有効な実験プロトコー ルを確定することができた。8週齢ルイスラット(180―220g)に大腸菌由来リポ多糖(LPS)200 μg を 300μl の溶媒に希釈して皮 下に注射。エキノクロームはLPS 投与と同時に PH7.4 の PBS を溶媒として 10, 1, 0.1 mg/kg(体重)を試験投与量とした。 EIU誘導24時間後にラット前房水内の炎症細胞数を計測すると、エキノクローム投与 10, 1, 0.1 mg/kg 群ではそれぞれ 115, 198, 30 80 千/ml であった。エキノクローム非投与群(陽性対象)では 2880 千/ml であった。無処置正常ラット前房水内(陰性対象)に は炎症細胞はみられなかった。エキノクロームは有意にラット急性ぶどう膜炎モデルを軽症化し(p<0.01)、その効果は濃度依存的で あった。
続いて前房水中の炎症性サイトカイン TNF-alpha の濃度も測定した。炎症細胞数の結果と同様にエキノクローム 10, 1 mg/kg 投与群 で有意に TNF-alpha 濃度が抑制されていた。 さらに NF-kappaB 核内移行、酸化ストレスシグナルも有意に減少することが明らかになり、英文原著論文として発表した(Mol Vis 20: 171-177, 2014)。
