| Outline of Annual Research Achievements |
In the first year of this project we sampled branchlets of Tsuga sieboldii and Tsuga diversifolia for genetic analyses along four elevational clines in Shikoku (Mt Ishizuchi area, Mt Higashiakaishi, Mt Shiraga and Mt Tsurugi). Branchlets of all samples have been scanned and photographed for morphological analyses. In addition all samples have had their DNA extracted. Core samples were also taken from all four sites and dendrochronological analyses indicate little or no correlation between trunk diameter and tree age.

3: Progress in research has been slightly delayed.
Reason
Obtaining permission to sample took longer than expected meaning that sampling took place in early November of 2024. This has slightly delayed the project meaning that results of genetic analysis won't be known until the second half of 2025.

| Strategy for Future Research Activity |
Genetic analyses (RAD-seq and cpDNA sequencing) will be undertaken in the second year of the project to identify the location of overlap zones of the two species and reveal the extent of hybridisation including backcrosses. If hybrid zones are identified we will revisit the sites and sample more extensively to clarify the spatial extent of each hybrid zone and genotype more samples to understand the hybridisation dynamics between the two species in more detail.We will also assess the coring data obtained in 2024 in more detail to see if coring of more trees is required.

| Causes of Carryover |
Genetic analyses could not be done in the first year so expenses were less than expected. The unused amount will be used in the second year and contribute to the cost of genetic analyses.
