Research Abstract

本年度は, 分子スケールおよび細胞スケールの統合ナノバイオメカニクスを主な研究課題として推進した. 得られた主要な成果は以下のとおりである.
(1)バクテリアの分離技術は細胞操作や水の浄化等で重要な基盤技術である. 流体力学, 細胞生物学, 生物物理学の知見を用いて, 流路内でのバクテリア挙動の理論・計算解析, MEMS技術による微小流路作成, PIV法による挙動計測の手法を統合し, バクテリアを遊泳能によって分離するマイクロチップを開発した(Ishikawa et al., Lab. Chip, 14, 1023, 2014).
(2)マラリア感染赤血球と血管内皮細胞に発現するタンパク間のリガンドーレセプタ結合の計算モデルを構築し, マラリア感染赤血球の細胞力学モデルを構築した. さらにこの手法をがん細胞の流動や血小板血栓形成の解析に応用した(Kamada et al., Microvasc. Res., 89, 95, 2013).
(3)クライオ電子線トモグラフィー法を用いて気管繊毛の三次元構造を解明した. また, マウスの気管上皮細胞を対象とし, 共焦点micro. PIVシステムを用いて繊毛細胞の作り出す流れ場を解明した(Kiyota et al., Am. J. Physiol. Lung Cell. Mol. Physiol., 306, L144, 2014)
(4)赤血球の二体干渉を精密にモデル化し, 細胞スケールの運動が血液のマクロなレオロジー特性に与える影響を解明した(Omori et al., Phys. Rev. E, 89, 013008, 2014 ; Omori et al., Comput. Mech., in press).
(5)繊毛を有する微生物が気液界面に捕捉される現象を発見した. またこの現象が走化性や走地性などの生物学的要因によるものでなく, 物理学的要因によるものであることを示した(Ferracci et al., PLoS One, 8, e75238, 2013).

1: Research has progressed more than it was originally planned.

Reason

研究は当初の計画以上に進展している. 平成25年度は, 分子スケールおよび細胞スケールの統合ナノバイオメカニクスを主な研究課題として推進し, マラリア感染赤血球の計算モデルを構築し, また, 繊毛の分子構造および繊毛細胞による流れ場を解明した. さらに統合ナノバイオメカニクスの最終目標である, 理論・実験・計算の統合的成果として, バクテリア分離マイクロチップの開発に早くも成功するなど, 計画を遥かに超える速度で研究が深化している. 研究成果は既に, 査読付き雑誌論文7編, 国際会議論文として16編発表されている. さらに, 医工学や計算力学の国際会議など招待講演を多数行い, またアジア太平洋地域最大の生体力学の国際会議で賞を受賞するなど国際的に評価されており, 当初の計画以上に進展していると評価できる.

Strategy for Future Research Activity

当初計画どおり, 平成26年度は引き続き, 分子スケールおよび細胞スケールの統合ナノバイオメカニクスに関する研究課題を推進する. 特に以下の項目を重点課題とする.
(1)マラリア, 血栓症, がんの血行性転移の数理モデル・計算モデルの構築および実験による検証.
(2)繊毛細胞による流れ場の数理モデル・計算モデルの構築.
