新皮質振動の錐体細胞サブタイプ依存性

Research Abstract

新皮質の電場電位には徐波、スピンドル波、ガンマ波といった多様な振動が観測されるが、その生成機構や機能分担には不明なことが多い。前頭皮質には投射先が異なる多様な錐体細胞があり、投射サブタイプごとに、各振動現象への関与が異なる可能性が考えられる。本研究では、錐体細胞投射サブタイプの電気的性質の違いや振動現象中の発火様式の違いを調べることで、錐体細胞と振動現象、それぞれの多様性の意味を考えたい。本年度は、in vitro標本で投射先を同定した錐体細胞の発火様式を調べるとともに、in vivo標本での錐体細胞の投射先同定法・形態解析のための細胞内染色法を確立した。
1. in vitro標本での、錐体細胞投射サブタイプの発火様式解析
背側前頭皮質の第5層で、対側皮質投射細胞の中で、同側の他の皮質領野に投射する錐体細胞の脱分極通電に対する発火応答を比較解析した。一次運動野に投射するものの発火様式は多様であったが、後部頭頂皮質や嗅周皮質36野に軸索を出すものは、比較的一様な応答を示した。これら投射サブタイプのin vivoでの発火様式を解析するのに基礎となる知見を得ることができた。
2. in vivo細胞外記録での錐体細胞サブタイプ同定と、それらの傍細胞記録による細胞内染色
背側前頭皮質第5層の橋核投射細胞、対側線条体投射細胞を、それぞれ橋核、対側線条体の電気刺激による逆行性応答で同定した(平均潜時:橋核投射細胞 6 ms;対側線条体投射細胞、18 ms)。傍細胞記録による細胞内染色の結果、橋核投射細胞は対側線条体投射細胞に比べて、1層での尖端樹状突起タフトが発達し、2/3層では逆に樹状突起側枝の数が少なかった。In vivoで錐体細胞の投射サブタイプごとに生理・形態解析できるようにしたことは、今後の皮質振動現象の解明に有用と考えられる。

新皮質振動現象を解析するのに基本となる、in vivoでユニット記録した錐体細胞のサブタイプ同定と記録細胞の細胞内染色法を確立できた。

Strategy for Future Research Activity

今年度確立した、in vivoでの錐体細胞サブタイプの電気生理学的・形態学的同定に基づき、それらのUp状態での発火様式を比較解析することを目指す。
