Outline of Annual Research Achievements

第二世代生理活性脂質として注目を浴びているリゾリン脂質は，細胞表面の特異的受容体を介し，多彩かつ重要な（病態）生理学的反応に関与する．しかし，生体におけるその変動・制御機構に関しては不明の部分が多く，臨床検体を用いたその測定の診断医学的応用も進んでいない．本研究においては，リゾリン脂質の（病態）生理学的意義・生体における動態のさらなる解明，関連物質を含む測定の臨床検査医学的応用，そして，その統合を通じ，「リゾリン脂質の病態検査学の確立」を目指した．
平成27年度は，我々のこれまでの生理活性脂質，リゾリン脂質に関する基礎・臨床的研究を集大成できるような成果を挙げることができた．
・スフィンゴシン1-リン酸（S1P）のキャリア・生物活性規定因子としてのアポリポタンパク質（ApoM）：LDL受容体/ApoEを介したApoM結合S1Pの代謝，ApoMを介したレスベラトロールの作用を明らかにすることができ，S1PのサロゲートマーカーとしてのApoMの重要性を提唱できた．
・リゾホスファチジン酸（LPA）産生酵素としてのオートタキシン（ATX）：世界で初めてATXアイソフォームの自動測定系を確立し，臨床検体の測定結果を公表した．現時点ではATXアイソフォーム測定の診断的意義は明らかにできていないが，今後の発展が期待できる．子癇の時の血清ATX低下が胎盤でのATX産生の低下によること，糖尿病腎症においてATXがバイオマーカーとなりうることを明らかにした．
・リゾホスファチジルセリン（LPS）の動脈硬化への関与を探るため，LPSのマクロファージに対する効果を検討した．泡沫細胞形成に対する促進作用，炎症に対する抑制作用というLPSの二面性が明らかになった．
