Outline of Annual Research Achievements

コンブ目の大きく性質が異なるそれぞれの科についての網羅的cDNA塩基配列情報の取得，多遺伝子解析による属・科レベルの詳細な系統関係の解析，大型化・深所適応の鍵となるいくつかの重要形態形質を，微細構造とそれらに関係する遺伝子群の解析から明らかにし，コンブ目植物が多様化し，世界各地で海の森を成立させるに至った進化の道筋を明らかにすることを目的として実施した。葉緑体６遺伝子（atpB, psaA, psaB, psbA, psbC, rbcL），ミトコンドリア６遺伝子（cox1, cox3, nad2, nad4, nad5, nad6）のDNA塩基配列に基づく多遺伝子系統解析を行い，各系統群の分布拡大時期と主要形態形質の獲得時期の推定を行った。また，昨年までに得られたトランスクリプトデータの詳細な比較解析を行い，細胞外タンパク質（secretome）のゲノムワイド解析から細胞外マトリクス再編成関連タンパク質の組成を明らかにし他結果，アルギン酸合成に関係するマンヌロン酸c5エピメレース，メタロプロテアーゼなどの酵素においてコンブ目の科レベルの進化に伴う遺伝子数またはドメイン構造の多様化が示唆された。また，プラズモデスマータ関連微細構造に関して，コンブ目植物と同様に複雑な多細胞体制を有するヒバマタの接合子の発生について観察をしたところ，数回の分裂を終えた時点ではPDは見られなかった。その後，最初の分裂面を含めて全ての隔壁内にPDが形成されていることが明らかになった。分裂を繰り返し多細胞となっているものの組織未分化の状態では，PDは隔壁内に分散して存在していた。ピットフィールドの出現は組織分化に伴って確認され、PFの周辺では既存のPDから分枝して新たなPDが形成される様子も観察された。二次原形質連絡の形成，PFの出現は，発生プログラムに合わせて行われていることが示された。
