Research Abstract |
All living creatures are made of cells-small membrane-bounded compartments filled with a concentrated aqueous solution of chemicals (A. Bruce et al. 1994). The relationship between the different water structures in the cells and its functionality has not been described yet. Our objective for the first year is to develop measurement and statisticalevaluation protocols to determine the water structure of somatic and callus rice cells. Measurement protocols were developed with two different instruments. The short wavelength NIR and NIR spectra of the cells were acquired in both transmittance and reflectance mode. Somatic and callus rice cells were monitored many times. Multivariate statistical evaluation processes and database manipulation methods were implemented using the program R-project. The data of the future experiments can be processed by the developed software automatically. The programs give the opportunity to build a database. The growing age of the somatic and callus rice cells can be predicted by their NIR spectra. The callus rice age prediction showed lower correlation (R2=0.727) and low prediction error (RMSEP=1.899). Somatic rice age prediction was better (R2=0.821, RMSEP=3.211). The regression models showed non linearity. The nonlinear growing characteristic may occur because of the water structural changes during the cell development. Many previously published water bands were assigned in the regression vectors of the prediction models. However, further analyses are necessary to assign the bands activated in the different stages of the cell development.

2: Research has progressed on the whole more than it was originally planned.
Reason
After the first year we have stable knowledge in the field of plant cell measurement by NIR spectroscopy using the Aquaphotomics approach. The achieved results are well agree with our previous plans and have scientific importance. We successfully developed measurement protocols and statistical evaluation process, which can used in the future. We started to build a database containing short wavelength NIR and NIR spectra of somatic and callus rice cells. This database with the developed software can be easily extended in the future to become more robust. In the field of somatic and callus rice cell development there is no agreement in the references concerning the number of stages. Our results can be used to define the main stages of the cell development during the growing of rice. The water bands found in the prediction model to predict the growing age of the rice shows the importance of water in the cell development. The water structure at the different stages of the cell development have to be further investigated. Therefore, the achieved results important not only at the present stage, but they are very promising for the future and the findings can give the basis of further researches. The results of the last year are appropriate for presentation at different national and international conferences and for publication in different interdisciplinary scientific journals. Furthermore, one of the most important achievements in the cell project is that we have enough knowledge to plan the continuation and to see the outcome of the project having the results of the first year.

Strategy for Future Research Activity |
The final main goal of our project is to build a database which contains the water spectral pattern of the most various cell kinds. We are planning to continue the cell project in the above mentioned field and we want to analyze different Lactobacillus strains as well in the next year. In the field of somatic and callus rice cell project we will define the mainstages of the cell development in the next year. We are planning more analyses with independent repeated experiments to describe the water pattern of the different stages. It can provide the possibility to determine the functionality of the differently structured water species in the rice plant. We are planning experiments to describe the characteristic water structure of Lactobacillus strains responsible for probiotic manner by Aquaphotomics approach. Probiotic and non-probiotic Lactobacillus strains isolated from yoghurt or purchased from company will be used. The NIR transflectance spectra of the cultures will be acquired during cultivation. The programs developed in the first year will be used to evaluate the results. The database will be also extended with the results. The important water bands for the discrimination between the bacteria strains will be determined. In longer term we are planning to further extend the database with different cell kinds in the third year. Finally, having a robust database will offer the possibility not only to determine the water structure of the different cell kinds, but to determine the general functionality of the differently structured water species common in the cells.

事務補佐員の退職により、新しい人材を採用した。 所定労働時間が大幅に少なくなり、人件費が削減となった。 次年度に繰り越して人件費、物品費を主として支出の予定。
