Outline of Annual Research Achievements

ヒト乳がん細胞株におけるMUC21の発現について、mRNAの存在、糖鎖付加を受けたポリペプチド産物の存在、その細胞内外における局在をヒト乳がん細胞株について比較検討した。伸長した糖鎖を持つMUC21に結合するheM21AとheM21C、糖鎖付加がないか又は短小なTn抗原を持つMUC21を認識するheM21Dを用いて、これらのグリコフォームを持つMUC21が細胞表面で検出できるかどうかをフローサイトメーターにより追求した。乳がん細胞株には既に公表されているmRNAプロファイルでは全ての細胞株で発現が見られるが、細胞表面にはいずれのタイプのMUC21も検出できなかった。しかし、同じ細胞をスライドグラス上に固定してこれらのmAbの結合性を見ると、細胞内におけるMUC21が検出され、MDA-231細胞ではゴルジ装置又は小胞体に局在している可能性が高かった。ヒト細胞株の由来となる乳がんのサブタイプと発現との関係に注目したが、現状では相関は不明であった。一方、MUC21の特定のグリコフォームの発現とアポトーシス耐性とが相関するという先行研究に基づき、糖鎖認識分子（ガレクチン３）の関与を追求した。細胞外ドメインの関与を明確にすることも目標に、HEK293細胞由来の全長MUC21トランスフェクタント、タンデムリピート部分を欠損するMUC21トランスフェクタント、及びモックトランスフェクタントにガレクチン３のsiRNAを導入し、ガレクチン３の発現低下を確認した（テキサス大学MDアンダーソンがんセンターと共同研究）。一方、多様な組織およびそれら由来のがんにおけるMUC21グリコフォームの発現の有無を、組織アレイを用いて追求した。mAb heM21Dの結合性は一部の扁平上皮由来のがん組織及び非がん病態組織に見られた（テキサス大学MDアンダーソンがんセンターと共同研究）。

Causes of Carryover

聖路加国際大学から順天堂大学への移動に伴う研究環境の構築は時間がかかり、一部予定していた研究内容の達成に遅れが生じた。

研究内容に変更はなく、資金の使途は後半から予定していた通りである。
