| Outline of Annual Research Achievements |
ナイーブマウスにWNTアゴニストであるR-Spondin1を3-6日間投与することで小腸におけるパネート細胞の増殖効果がみられることが明らかとなり、R-Spondin1がパネート細胞増殖活性を有することが証明できた。さらに陰窩と腸絨毛間に、パネート細胞と杯細胞の中間の形質を有するintermediate細胞の出現が新たにみられた。本細胞はMMP7陽性、defensin陽性であり、抗菌活性を有していた。その意義について解明を今後進めている。また大腸には異所性パネート細胞の出現が観察された。一方、臨床のGVHD患者の大腸内視鏡生検病理標本でに異所性パネート細胞がみられることを明らかにし報告した（Shimoji, BMT 2013)）。これらの結果から疾病によって大腸に異所性パネート細胞がみられるメカニズムにR-spondinの産生亢進が関与する可能性が示唆された。そこで、GVHD発症時のR-spondin産生細胞を同定する研究を開始し、マウスでは小腸の間葉系細胞で発現がみられることが明らかになった。今後、R-spondinを産生する細胞群の同定と、その産生調節にかかわる分子メカニズムの解明を行う。また、われわれは糞便中抗菌ペプチドdefensinのELISA測定系を開発することができ、この測定系を用いて、GVHD発症後のパネート細胞からの腸管内腔へのdefensinの分泌の減少を確認した。さらにR-spondin投与により腸管内腔へのdefensinの分泌が回復することを世界に先駆けて証明でき、今後その産生の経時的変化を含め、詳細な解析を行う予定である。
