核小体の形成メカニズムと栄養飢餓応答における役割

Outline of Annual Research Achievements

真核生物の細胞核は、遺伝情報の本体であるゲノムDNAを包括して、その構造や機能を制御する空間である。核小体は細胞核内の最大の構造体で、リボソームが生合成される場である。リボソームRNA 遺伝子の反復配列を含む染色体領域を中心に形成され、中心から辺縁にむかって順にrRNAの転写、プロセシング、リボソームのアセンブリーが行われる。核小体で構築されたリボソームは、その後核外に輸送され、細胞質におけるタンパク質一般の産生に機能する。リボソーム合成はrRNAの大量の転写をはじめとして、細胞内のエネルギーを大量に消費する過程であることから、核小体は細胞のエネルギー恒常性の維持や、栄養飢餓応答や細胞増殖制御などに密接に関わるとされているが、詳細な分子機序は不明である。本研究では、核小体の形成・維持に関わる因子を検索、同定し、作用機序を解明することで、核小体の本態を明らかにすることを目的とした。まず、siRNA ライブラリーを用いたハイコンテントスクリーニングをもとに、ノックダウンすることで核小体の形成に異常を示す、いわゆる“核小体ストレス”の状態を引きおこす因子を15程度同定した。これらのノックダウンが誘導する核小体形態を、高度画像解析技術を用いてプロファイリングし、３種のタイプに分類できること、また、一群のリボソームタンパク質が減弱すると、共通した核小体異型を示すことを定量的に示した。また、pre-rRNAの合成、rDNAの遺伝子配置、細胞増殖などに影響があることや、これらの細胞表現はタンパク質の翻訳阻害による２次的なものではないことを明らかにした。以上のことから、リボソームタンパク質の持つ新たな核小体の構造タンパク質としての機能を見いだした。
