Outline of Final Research Achievements |
Hepatic peroxisome proliferator-activated receptor γ (PPARγ)-dependent gene 1 (HPD1) gene was isolated as high expression in fatty liver and unknown function. The HPD1 gene is directly regulated by PPARγ. In the present study, to assess physiological function of HPD1 protein, HPD1 expression was repressed by lentivirus expressing HPD1shRNA. Infection of lentiHPD1shRNA dramatically decreased HPD1 protein by about 20% as comparison with lenticontrolshRNA. Although normal or phospho-specific antibody array was performed using lenticontrol and HPD1shRNA-infected cells,there was no significant difference between the protein or phosphorylation levels of both cells. To assess subcellular localization of HPD1 protein, 3T3-L1 adipocytes were fractionated by differential centrifugation. The result showed that the HPD1 is exclusively detected in the microsomal fraction. Interestingly, HPD1 localized in detergent-insoluble microsomal fraction.
