ヒト純化細胞を用いた肝多段階発がん過程におけるエピゲノムプロファイル解析

HBV陽性・HCV陽性肝細胞がん症例の背景非がん肝組織からそれぞれ肝細胞を純化し、全ゲノムバイサルファイトシークエンスならびにH3K4me1・H3K4me3・H3K27Ac・H3K36me1・H3K27me・H3K9me1の6種類のヒストン修飾に対するChIP-seqを行った。
純化正常肝細胞の標準DNAメチル化プロファイルと比較すると、ウイルス感染を伴う肝細胞では正常肝細胞における個体差を遥かに凌駕するDNAメチル化変化があった。ウイルス種によるDNAメチル化変化の差異を比較すると、HBV感染肝細胞に比してHCV感染肝細胞ではより多くのDNAメチル化変化が蓄積していた。HBV感染肝細胞ではDNAメチル化亢進が、HCV感染肝細胞ではDNAメチル化減弱が目立っていたが、HBV感染肝細胞においてもCpGアイランドではDNAメチル化亢進が優位であった。個体差が少なく、DNAメチル化の変化から何らかの機序で保護されているとみられる転写開始点から200bp以内や第1エクソンにおいても、HCV感染肝細胞ではDNAメチル化の変化がみられ、保護機序の破綻が推察された。
組織検体を用いたInfiniumビーズアレイ解析において、HBV陽性肝に比してHCV陽性肝ではDNAメチル化異常の頻度が高く、DNAメチル化亢進の頻度が高い結果が得られており、純化肝細胞の解析結果はこれと一致していることが確かめられた。
組織検体のInfiniumビーズアレイ解析データより、HCV陽性肝細胞がんにおいて前がん状態より生じがんに受け継がれるDNAメチル化異常を示すCpG部位を特定し、その部分のヒストン修飾状態を純化肝細胞によるChIP-seqで評価した。同部位においてはH3K4me1とK27Acがより集積しており、肝発がんに寄与するDNAメチル化異常に先行するのはK4me1+K27Acのactiveマークの可能性があると考えられた。
