Outline of Annual Research Achievements

免疫組織におけるIL-15産生細胞の分布と機能を明らかにするために、IL-15-CFPノックインマウスを作製し、IL-15産生細胞の生体内分布を調べるとともに、組織特異的IL-15遺伝子破壊（IL-15cKO）マウスを作製して、各組織におけるIL-15の機能を明らかにする。
IL-15cKOマウスの作製については、Foxn1-Cre、Villin-Cre、Alb-Cre、Tie2-Cre、CD11c-Cre、MuCre-A、LysM-Cre、Prx1-Cre、Adipoq-CreなどのCreマウスとの交配が順調に進んでいる。Villin-Cre、Alb-Cre、Tie2-Cre、Adipoq-CreマウスについてはIL-7-floxマウスとも交配し、IL-15/IL-7ダブルcKOマウスも作製中である。
IL-15cKOマウスの解析については、Foxn1-Cre、Alb-Creについての解析が進行中である。Foxn1-Cre IL-15cKOマウスの胸腺では、NKT細胞とγδ T細胞が減少していた。特にNKT細胞についてはより成熟した段階のものの減少が大きかった。これらの結果はIL-15欠損マウスの表現型とよく一致しており、胸腺の中では胸腺上皮細胞が産生するIL-15が主要な働きをしていることが示された。

Reason

IL-15cKOマウスの作製については、Foxn1-Cre、Villin-Cre、Alb-Cre、Tie2-Cre、CD11c-Cre、MuCre-A、LysM-Cre、Prx1-Cre、Adipoq-CreなどのCreマウスとの交配が順調に進んでいる。また、Villin-Cre、Alb-Cre、Tie2-Cre、Adipoq-CreマウスについてはIL-7-floxマウスとも交配し、IL-15/IL-7ダブルcKOマウスも作製中である。IL-15cKOマウスの作製については、Foxn1-Cre、Alb-Creについての解析が進行中である。以上のことから、ほぼ満足できる達成度と判断した。

Strategy for Future Research Activity

IL-15cKOマウスの解析は、引き続きCreマウスとの交配を進めるとともに、平成27年度中に複数のCreマウス系において解析を進めていく。また、Tie2-Cre IL-15cKOマウスでは、多光子顕微鏡を用いて生体内でのリンパ球の動態に変化がないかを解析する。

Causes of Carryover

国内学会発表のために使用予定であった旅費50,000円を他の予算から支出したために余剰が乗じた。

次年度使用額64,818円は、翌年度分の助成金とともに、主に物件費として使用する計画である。
