原虫感染に対するCARD9を介した新規自然免疫活性化経路の解析

Outline of Annual Research Achievements

本研究では、寄生虫のうち、原虫を認識する自然免疫受容体とそのリガンドの同定を目指した。前年度までに、ITAM受容体の下流でアダプター分子として機能するCARD9に依存する自然免疫活性化経路が原虫Leishmania major感染に対する抵抗性に関与することを確認した。今年度は、CARD9の上流に位置するSykキナーゼ欠損マウスを用いて、同様の結果を得、この経路がL. major感染抵抗性に関与していることを確認できた。CARD9の上流に位置する受容体に関しては、ITAM受容体の可溶型蛋白ライブラリーを用いて、認識受容体の候補を得た。これらの原虫への結合は確認できたが、認識するリガンドの同定には至っていない。現在、リガンドの同定を目指すとともに、この受容体の自然免疫活性可能、およびTh1型免疫誘導能などを解析している。
