Outline of Annual Research Achievements

本研究は骨髄由来抑制細胞(MDSC)をターゲットとした新規悪性黒色腫治療法の開発を目的として、マウスと人の細胞を用いて研究を行った。本研究により、腫瘍内でMDSCから分化する抑制型マクロファージである腫瘍随伴性マクロファージ(TAMs)が、インターフェロンベータにより成熟過程で抗腫瘍型マクロファージに分化することを明らかにした。また、同時にインターフェロンβが腫瘍内浸潤リンパ球上のPD-1の発現を上昇させるため、抗PD-1抗体を併用したところ、マウス悪性黒色腫モデルにおいて、強い抗腫瘍効果をもたらすことが明らかになった。これを臨床応用するため、ヒト単球由来マクロファージを用いて、インターフェロンβがマウスと同様にケモカインプロファイルを抗腫瘍型に変化させることを明らかにした。以上をOncoimmunology (IF 6.266)に掲載した。さらに臨床応用を目指し、現在、上記研究を医師主導臨床研究として、東北大学病院倫理委員会の承認のもと、phase 1b研究を行っている。他に、乳房外パジェット病におけるTAMsの役割の検討をJournal of Investigative Dermatology (IF: 7.216)に2報、Experimental Dermatology(IF:3.762)に1報、菌状息肉症におけるTAMsの役割の検討をExperimental Dermatology(IF:3.762)に1報、報告している。
