環境因子と皮膚老化・皮膚疾患‐AhRを中心とした分子メカニズム解析

Outline of Annual Research Achievements

Aryl hydrocarbon receptor(AhR)を中心とする機序解析から、皮膚老化、皮膚疾患との関連や、TH17系の免疫におよぼす作用を行っている。まず、分子疫学的調査:人種差による環境因子(紫外線・喫煙・大気汚染)対する皮膚老化の差違―JAGE project(Study of extrinsic skin ageing of Japanese and German women)のー大規模調査(ドイツ側200名・日本側200名)として、皮膚老化スコア(SCINEXA)を使用し、新たな質問表(Life style, Food, Sun exposure, Womenの4種類)を作成し、名古屋市内(近傍)100名、大気汚染の少ない地域で100名の調査を行った。調査は、終了し解析を進め、共同研究者であるドイツデュッセルドルフ大学環境医学研究所には血液サンプルを送付した。また、タバコ抽出液によるAhR経路の活性化についても、角化細胞、線維芽細胞ではコラゲナーゼの誘導、さらに、メラノサイトでもMITF発現上昇、メラニン産生の増加を見いだした。また、紫外線やタバコの喫煙が関与する乾癬や掌蹠膿疱症の患者においても、単一ニクレオチド多型(SNPs)解析を行った。

Reason

重要な分子疫学調査は終了し、引き続き、ドイツデュッセルドルフ大学環境医学研究所とともに、解析を行っているところであり、他のアジア(中国)での解析とあわせ、人種差以外の環境因子にもさらに焦点をあてて解析をすすめている。全容の解析には、さらに今年度内、時間がかかるものと思われる。

Strategy for Future Research Activity

分子疫学調査の解析を引き続きおこなうことと、さらには、紫外線やタバコの喫煙が関与する乾癬や掌蹠膿疱症の患者においても、単一ニクレオチド多型(SNPs)解析、TH17/制御性T細胞の解析もあわせて行い、環境因子とTh17/制御性T細胞の関係を明らかにする予定である。
