Outline of Annual Research Achievements

①膵癌細胞株より新たにGemcitabine耐性株の樹立。膵癌細胞株(MiaPaka2)をIC50値の濃度のGemcitabineで6ヶ月培養を行い、その中でGemcitabineに対する強い耐性を取得し、表現型の変化、浸潤能の増加を示す株を作成した。
②表現型や浸潤能に関与する遺伝子を同定した。樹立耐性細胞株と親株をDNA microarrayで比較検討すると、耐性株においてMAST4やPDE1の強発現、変異KRASシグナルの異常を同定した。
③３次元培養と２次元培養の遺伝子発現パターンを比較した。このMiaPaca2は３次元浮遊細胞では耐性化が起こり、２次元培養では致死量のGemcitabineの効果が減弱することがわかった。
