アンジオテンシン1-7による前立腺癌細胞のアンドロゲンレセプター発現調整の解明

Outline of Annual Research Achievements

AngIIからアンジオテンシン変換酵素２（ACE2）によって変換されるペプチドのAng1-7が抗腫瘍効果を有するとした基礎研究が報告されている。ACE阻害剤は、直接ACE2には作用しないが、AngI濃度を上昇させることによってAng1-7産生を促進したり、Ang1-7の分解を予防したりする間接的な作用を持つと推察されている。Ang1-7は、AngIIレセプターのAT1RあるいはAT2Rには結合せず、GPCRであるMasレセプターに結合することが分かっている。一方、アンジオテンシン２レセプターの一つであるAT2レセプターは、心血管系細胞においてAT1レセプターシグナルと拮抗作用があり、血圧降下作用や心血管系細胞の増殖抑制などがあるとの報告がある。従って、AT2レセプターのリガンドは降圧剤としての作用を持ち、現在グローバルで臨床試験が行われている。Ang1-7を介したMasレセプターシグナルも同様の作用が予想されている。
そこでAT2レセプターリガンドの前立腺癌細胞に対する作用について検討した。細胞増殖に対しては、濃度依存的に抑制することが分かった。これは、アンドロゲンレセプター（AR）の有無に関係なく、どの前立腺癌細胞でも抑制効果を示した。また、ARに対するC21の影響を調べたところ、アンドロゲン（DHT）が細胞質でARと結合して核内に移行するのを、C21は抑制していることが分かった。前立腺癌を発症するトランスジェニックラット（TRAP）を用いて、C21を継続投与して前立腺癌発症の影響について調べた。その結果、病理学的解析によると前立腺癌の発症は抑制された。以上の結果より、AT2レセプターリガンドであるC21は、前立腺癌発症を抑える可能性が示唆された。
