| Outline of Annual Research Achievements |
顎関節罹患者における睡眠呼吸障害の高い罹患率に着目し、呼吸障害における間欠的低酸素曝露と軟骨を介した顎骨成長との関連について検討するため、8週齢雄性SDラットを用いた間欠的低酸素曝露動物モデルにおける（１）顎骨成長、（２）関節頭部のリモデリングに関して、第一に形態学的およびCT解析を実施した。その結果、（１）生後7週齢から10週齢までの成長期における低酸素曝露群において、形態学的解析により、鼻骨および下顎骨の低成長が有意に生じること、さらに、（２）下顎頭軟骨層近傍において骨密度の上昇が認められた。軟骨層近傍における膜性骨化を成長様式の中心とする顎骨と、軟骨内骨化を成長様式の中心とする四肢骨との比較を行った結果、その影響を顎骨において、より顕著であることが判明した。さらに、顎骨とともに気道を構成し、低酸素環境に密接に関わる（３）鼻上顎複合体、（４）上部気道筋における形態および代謝系の変化に関して解析を行った。その結果、（３）鼻上顎複合体の多くを占める硬組織気道の体積の減少が生じ、鼻中隔軟骨およびその近傍組織を対象とした炎症及び成長関連因子の定量PCRから、eNOS（血管内皮型NOS)の発現低下が認められた。また、（４）上部気道筋である舌骨筋を対象に、炎症および細胞死の関連因子の定量PCRを行った結果、炎症系サイトカインであるIL-1β、TGFαおよびiNOS（誘導型NOS）の発現上昇が認められ、また、BNIP3の発現上昇から筋組織におけるオートファージ変性の誘導の可能性が示唆された。
