Outline of Annual Research Achievements

ロコモティブシンドローム(運動器症候群)の原因となるサルコペニア(加齢性筋肉減弱症)は,要介護や長期寝たきりの誘因となり,筋萎縮が起こる.そこで運動不可能時などにおける栄養療法による骨格筋と血管構造病変に対する修復再生方法を開発し,その機序を解明する.本研究は,寝たきりや可動域制限や術後など運動できない人のために,食品栄養成分による骨格筋・血管病変を改善する体内環境の構築を目的とする.運動と同様の体内環境を惹起する運動・メカニカルストレス様作用をもつ食品成分を分子レベルで探索・データベース化し,骨格筋とその血管の調節因子について解析を行う.高齢化社会にむけてエビデンスに基づく,食品成分による運動器の革新的刺激因子の検証・開発を行う.平成26年度はin vitroにて血管退行・筋萎縮改善の運動時メカニカルストレス様食品成分の候補を絞り込み,モデル動物を作製してこれらの経口投与を行った。微小血管構造異常・筋萎縮改善効果について,各因子の産生や遺伝子発現を検証した.骨格筋と血管について運動時の活性へ誘導する刺激成分を絞り込み,筋肉の材料としての栄養成分とは異なる,運動・メカニカルストレス様刺激作用を持つ成分を明らかにして食品成分から構成される栄養サポート法を考案する計画である.

Reason

平成26年度は主にin vitroにて血管退行・筋萎縮改善の運動時メカニカルストレス様食品成分の候補を絞り込み,モデル動物を作製してこれらの経口投与を行った。微小血管構造異常・筋萎縮改善効果について,摘出した骨格筋から各因子の産生や遺伝子発現を検証している。効果の測定について解析数が多いため,対象物質や解析の検討を行い研究を進める計画である.

Strategy for Future Research Activity

平成27年度においては本年度のデータベースを完成させるために微小血管構造異常・筋萎縮改善効果の測定について,解析パラメータを再検討して研究を進める計画である.また,筋萎縮モデル動物に投与するための成分の厳選を行う計画である.筋萎縮モデル動物を作製し、有効成分を投与し,血清から生化学検査を行い,下肢の筋肉組織など各組織を摘出し,組織切片を作製して組織化学染色,蛍光免疫組織化学染色,筋原線維タンパク質解析,血管構造解析を行う.これら三次元構造・生化学的・遺伝子発現分析から、運動器3D構造の改善について検証を進める計画である。
