光によるウイルス感染の検出・制御技術の開発

Outline of Annual Research Achievements

ラマン分光およびレーザー技術を基盤とし,ウイルスに感染した細胞のウイルス種類の判別やレーザー照射による細胞制御を可能とする技術の開発を行った。ラマン分光分析を用いると,無標識で無侵襲的に細胞の状態変化を解析することが可能である。本研究ではHEK293細胞内で増殖可能なアデノウイルスと増殖不可能な物を用いて,いかに早く検出が可能か,また,検出のメカニズムを明らかにするための研究を行った。
ウイルス感染によるラマンスペクトルの変化は非常に小さい。細胞を培養する際のわずかな変化,例えば培養液成分の変化や細胞継代数の違いなどによる変化と同レベルの変化である。しかし,実験を繰り返して統計的に解析することにより,培養環境による変化に重複しない,ウイルス感染による変化の特徴を捉えることに成功した。
現在の記録では,ウイルス添加後6時間で感染を検出することに成功している。免疫染色法を用いると,6時間後にはすでに核内の初期タンパク質の増加が観測されており,これがラマンスペクトル変化の原因と考えられた。しかし,増殖因子のうち二つを持たず,HEK293細胞内でも増殖不可能なアデノウイルスについても,感染の検出が可能であることが分かった。
増殖が可・不可のウイルスに感染した細胞のスペクトルを解析した結果,感染初期に生ずる変化と,感染後に生ずる変化の二つがあることが示唆された。これらは,細胞自身の反応と初期タンパク増加による影響の二つにそれぞれ帰属できる可能性がある。
