Outline of Final Research Achievements |
I have attempted to produce a transgenic medaka that express a cilia-related gene in a spatiotemporally regulated manner by combination of Transcription Activator-Like Effector Nuclease (TALEN) method, Cre/loxP system and heat-shock promoter. I have already achieved that a TALEN knockout medaka in which a cilia-related gene in knocked out. This fish line developed a polycystic kidney as expected. Cystic dilation was already detected at hatching and gradually increased in size as fish grew. This phenotype resembles that of human polycystic kidney disease. Construction of a transgenic line using kidney specific promoter in progress.
