| Outline of Annual Research Achievements |
本年度では、すでに作製されたアポEノックアウトウサギの解析を行い、以下の研究成果を得た。ホモ結合体のアポE欠損ウサギでは野生型ウサギと比べ血中の総コレステロールと中性脂肪の増加が見られた。また、血中のアポBが上昇しているが、アポAIの変化はなかった。超遠心法及びHPLC法により、リポタンパクの変化を検討したところ、アポE欠損ウサギには著しくレムナントリポタンパクの増加が認められた。これらのリポタンパクのアポB48の量がアポB100の量を上回っていることが確認できた。さらに、高脂肪食を2週間負荷させたところ、アポE欠損ウサギの総コレステロール値は1200 mg/dlまで上昇しており、高脂肪食に対する敏感性が非常に高いことが判明した。これらの結果から、アポE欠損ウサギは新しい高脂血症モデルになりうると考えられ、今後、動脈硬化研究にも応用できるかを見極めていきたいと考えている。現在はこれらの成果をまとめ、投稿中である。また、アポCIII欠損ウサギの繁殖を開始しており、ホモ結合体アポCIII欠損ウサギの獲得を目指している。
TALEN技術より作製されたアポAII遺伝子ノックインウサギはgermlineができず、Cas9技術により新たにアポAIIノックインウサギの作製を試みた。すでに出産した１７羽のウサギのうち、２羽がアポAII遺伝子のノックインがPCR法により確認された。今年の夏ころからの繁殖開始が期待される。

Reason
アポEとアポCIII欠損ウサギの作製と解析は順調に行われてきたが、アポAIIノックインの成功率が相当低い。それを克服するために、Cas9法を活用し、新たな技術で作製を行っている。

| Strategy for Future Research Activity |
今後は、アポCIII欠損ウサギの繁殖を行いながら、アポAIIノックインの成功を加速したい。また、アポCIII欠損ウサギの解析も早期に実現させたい。
