ウイルスを特異的に検出するためのハイブリッド誘電泳動インピーダンス計測法の開発

Outline of Annual Research Achievements

特定のウイルスを電気的に検出システムの開発を目的に研究を行った.当初目的では,ウイルスを誘電泳動によって微細電極に捕集し,その捕集に伴う電極間のインピーダンス変化を計測すること,さらに,ウイルスの捕集の際に抗原抗体反応を利用して,特異的に検出することを目指していた.
しかし,抗原抗体反応を利用したとしてもウイルスを高感度に検出することができなかった.それは,ウイルスによる電極間のインピーダンス変化が非常に小さいためである.
そこで,ウイルス遺伝子を増幅する核酸増幅法,具体的にはPCR(polymerase chain reaction)と利用することを考えた.PCRは,DNAを特異的に指数関数的に増幅する手法で,これを利用したウイルス検査は非常に感度および正確性が高い.一方で,PCRによって増幅されたDNAは何らかの方法で検出されなければならず,それは手間のかかる作業であった.
そこで,PCR後の増幅DNAを電気的に迅速かつ簡便に測定する手法を考案した.それは,増幅DNAを誘電体微粒子に結合させて,そのDNA結合微粒子を上述のように誘電泳動集積して測定する.このとき,適切な誘電体微粒子を選択することで,DNAが結合した微粒子のみを選択的に捕集できることを見いだした.即ち,検出対象DNAのみを選択的に測定できる.測定に必要な時間は,DNAと微粒子の結合に15分,電気的検出に10秒とこれまで1時間~3時間程度必要であったPCR後のDNA検出が15分程度で行うことができるようになった.
