抗原特異的遅発型喘息反応の生物学的、薬理学的検討

Outline of Annual Research Achievements

T細胞依存性喘息反応のメカニズム解明の目的に、OVA特異的T細胞クローン移入モデルを樹立した。平成25年度にはまず、OVA特異的T細胞受容体トランスジェニックマウス脾細胞より、多数のT細胞クローンを樹立した。さらに、無処置のBalb/cマウスにT細胞クローンを移入し、翌日OVAを経気道的にチャレンジした後、経時的に気道抵抗（RL）、呼吸パターン（Penh）を測定し、気流閉塞をモニターした。OVAあるいはT細胞エピトープペプチドOVA309-323チャレンジにより、RL、Penhの上昇を認めたことにより、T細胞依存性の遅発型喘息反応（アレルギー反応）の存在を証明できた。この反応のメカニズム、メディエータ解明に向け、培養気管支平滑筋を用いたin vitroの収縮活性アッセイ系を樹立した。マウスprimary気管支平滑筋細胞を培養し、90%以上の純度となった時点で、コラーゲンゲルに包埋し、さらに6日間培養、成熟させた後、収縮活性検出に用いた。in vivoモデルで気流閉塞を惹起するT細胞クローンを固相化CD3抗体で刺激し、その培養上清をKrebs液に透析後、ゲルにapplyした。無刺激のクローン上清に比較して、活性化T細胞クローン上清では、有意に平滑筋ゲルの収縮活性が確認された。montelucast (10 microM)との前培養では、ゲルの収縮に変化はみられなかった。本知見は、T細胞がロイコトリエンとは異なる気管支平滑筋収縮活性物質を産生していることを示す。一方、in vivoモデル解析においては、ロイコトリエン拮抗薬montelucast (0, 1, 5 mg/kg)を前投与したところ、Penh上昇が抑制され、T細胞依存性の気流閉塞は、ロイコトリエンの作用を介していることが示された。今後、このdiscrepancyについて、さらに解析を進めることが重要と考えられる。
