Outline of Annual Research Achievements

研究代表者は現在までに、IgG-Fc領域を特異的に表面に固定化出来るバイオナノカプセル (ZZ-BNC)を開発し、抗体分子のクラスター化と整列化、更に固相上に整列化された抗体を検出する能力を有することを見出し、各種イムノアッセイやイムノセンサーを高機能化すると共に、抗体分子の整列化度合いを評価するプローブを開発した。そこで本研究は、同技術を抗体以外のセンシング分子にも応用し、バイオセンサー全般の飛躍的な機能向上を目的とした。
最終年度は、H26年度に見出した水晶発振子微量天秤（QCM）センサーを用いたZZ-BNCによるレセプター-リガンド相互作用の高感度化技術の汎用性を高めるために、表面プラズモン共鳴（SPR）センサー、酵素標識リガンド結合アッセイへ展開し、両手法の検出感度及びリガンド結合能が著しく上昇することを見出した。さらに、ZZ-BNCの平面膜化を検討し、より広範なバイオセンサーの高感度化、高機能化を図っている。なお、これらの成果は、原著論文、学会、ホームページ等で発表し、社会・国民に向けて積極的に情報発信を行った。
本研究期間全体を通じて、まず、Protein GおよびProtein L由来の各IgG結合ドメインをもつBNC(LG-BNC, LL-BNC)を作製し、ZZ-BNCでは結合力が弱いあるいは結合しない抗体と結合能を有することを見出し、優位性を示した。さらに、抗体以外のセンシング分子（Fc融合レセプター）に応用展開し、QCM、SPR、酵素標識リガンド結合アッセイのリガンド-レセプター相互作用の検出感度を飛躍的に向上することに成功した。本研究期間に得られた成果は、原著論文、特許申請、学会、ホームページ等での発表を行い、社会・国民に向けて積極的に情報発信を行ったことから、本研究の意義、重要性は非常に高いと考えられる。
