Periodontitis is a choronic inflammatory disease accompanied by alveolar bone resorption by osteoclasts. Porphyromonas gingivalis, one of the major pathogens of periodontitis, produces cysteine proteases called gingipains, that are clasified into Rgp and Kgp based on their cleavage site specificities. We previously found that Kgp degrades osteoprotegerin (OPG: 56kDa), an osteoclastogenesis-inhibitory factor, into an N-terminal fragment (37kDa) and inactivates OPG. In this study, The principal investigator tried to determine the specific cleavage sites in OPG cleaved by Kgp and prepare mutant OPG proteins resistant to the cleavage by Kgp. Unfortunately, a mutant OPG protein resistant to Kgp has not been obtained.
