金属ナノ粒子による細胞内分子イメージング

Outline of Annual Research Achievements

本研究計画では、細胞内分子イメージングのための「3次元・ナノ・分析・光学」の顕微鏡開発を目的とし、昨年度には下記の4つの項目を達成した。
1、細胞内3次元追跡の高速化、マルチプローブ計測の実現
細胞内の金属ナノ微粒子を追跡しながら周囲の分子をセンシングする技術の向上に取り組んだ。様々な形状・種類のナノ微粒子の合成と、励起波長の最適化により生きた細胞内部で1msの時間分解能と30nmの位置精度でのナノ微粒子の3次元追跡及び表面増強ラマン散乱(SERS)分光計測を達成した。また、複数の金属ナノ微粒子を細胞内で同時に追跡・SERS分光計測が可能な顕微鏡の開発にも成功した。
2、機能的・高感度な金属ナノ微粒子プローブによる細胞内イメージング
開発した高感度で安定なSERS検出を可能とするコア-サテライト状金属ナノ微粒子をp-MBAにより修飾し、pHセンシング機能を付加した。修飾した金属ナノ微粒子を細胞内に導入し、アポトーシス時の局所領域でのpHを経時的に測定した。アポトーシスの進行による細胞形態の変化に伴いpHの変化が検出された。細胞内環境変化を金属ナノ微粒子を用いて検出することに成功した。
3、薬剤取り込み過程のリアルタイム計測
細胞の3次元イメージングが可能なラマン散乱顕微鏡を開発し、薬剤を付加した細胞の経時観察を行った。細胞へは金属ナノ微粒子を事前に取り込ませ、薬剤が細胞内に到達した場所と時間を、薬剤に付加したアルキンのSERSにより検出した。細胞への薬剤の付加後、数十分での薬剤取込・取込量と薬剤濃度との相関・この薬剤の取り込みにはエンドサイトーシスが関与しないことを確認した。
4、深紫外領域での応用を目指した金属ナノ微粒子の合成と評価
プラズモン共鳴波長が深紫外域となるインジウムナノ微粒子の合成をした。インジウムナノ微粒子を塗布した石英基盤上にアデニンを付加し、生体分子の深紫外SERSを確認した。インジウムナノ微粒子による局所増強効果を利用した生体内での深紫外ラマン分光計測実現の可能性が示唆された。
