気孔装置解析による植物独自の高次情報処理のパラダイム提案

Outline of Annual Research Achievements

（１）気孔機能化因子SCAP1転写因子の調節機構の解明：気孔の分化機構は知られているが、分化過程終了後に始まる気孔環境応答能獲得の機構は未解明である。我々はDOF型転写因子SCAP1が気孔を成熟させ、機能化するための司令塔であることを明らかにしたが、その発現調節機構には手つかずであった。そこで、このSCAP1遺伝子のプロモーター領域で発現制御に関与する領域の決定を行った。引き続き酵母ワンハイブリッド系を用いて特異的にSCAP1に作用する転写因子の同定を試みているところである。
（２）気孔閉鎖因子SLAC1陰チャネルにおけるABA・CO2情報感知・処理機構の解明：SLAC1の活性化制御に関しては、ABAシグナルによりN末端領域がリン酸化されて活性化するとの報告はあるものの、CO2によるSLAC1の活性化に関しては何ら報告されていない。変異型SLAC1遺伝子を導入した形質転換植物の解析から、SLAC1のCO2シグナル受容部位はそのチャネルタンパク質の膜貫通領域を形成する部分に存在することを明らかにした。さらに、CO2シグナル受容に関与するアミノ酸を同定するため、SLAC1の立体構造モデルを基にリン酸化されうるアミノ酸を選抜し、1アミノ酸置換実験を行った結果、CO2応答に特異的に関与すると思われる２つのアミノ酸残基を同定した。

Reason

気孔環境応答における高次情報処理を司る因子として、気孔機能化因子SCAP1と気孔閉鎖因子SLAC1チャネルの二つを中心に研究を進めている。どちらも、植物細胞の多元的信号処理の要の因子である可能性が高い。前者においては新規調節因子の同定が進んでおり、後者においては環境刺激の違いを異なる受信部位によって選別していることを明らかにした。研究題目に合致した成果が今後得られることが期待される。

Strategy for Future Research Activity

２年次以降の研究においては、初年度で得られた研究成果を踏まえ、環境情報の統合に関する細胞レベルでの分子機構の解明につなげていきたい。また、高次情報処理の新規事例として、孔辺細胞と副細胞からなる気孔装置にも着目し、２つの細胞間の協調行動に介在する膜交通機構の解析から、細胞間コミュニケーションについての研究も展開する予定である。
