階層を貫く細胞サスペンジョン力学の新展開

Outline of Annual Research Achievements

本年度は、ナノスケールからマクロスケールまで、幅広い研究課題で当初の予定を上回る成果があがった。
気道の繊毛流れに関する研究では、気道繊毛の分子モーターの構造を解明し(Ueno, et al., Cytoskeleton)、この情報を基に、繊毛運動を再現できる数値シミュレーションコードを開発した。さらに、繊毛が作り出す流れに及ぼす粘液粘度の影響を実験的に明らかにし、気道病理の解明に役立つ基礎知見を得た。微生物懸濁液に関する研究では、鞭毛の協調運動メカニズムの解明(Kanehl & Ishikawa, Phys.l Rev. E)や、懸濁液内の拡散特性(Ishikawa & Pedley, Phys. Rev. E)まで、複数のスケールにまたがる研究成果をあげた。カプセルに関する研究では、流れ中の単一カプセルの移動現象を解明し(Nix, et al., Phys. Rev. E)、大規模懸濁液のレオロジー特性を明らかにした(Matsunaga, et al., J. Fluid Mech.)。血流に関する研究も順調に進展し、単一赤血球の運動挙動(Omori, et al., Comp. Mech.)や、小血管内の軸集中現象の解明(Takeishi, et al., Physiol. Rep.)等の成果をあげた。微小流体流路に関する研究も進展し、細胞スケールからcmスケールまで高解像度で計測する新しい技術を開発した(Kawano, et al., PLoS ONE)。さらに、細胞サスペンジョン力学を臨床応用することを目的に、ナノ酸素バブルを含む輸血用懸濁液を開発した(Matsuki, et al., J. Nanomedicine)。こうした応用研究は、最終年度に実施する予定であったが、研究の進展が当初の予定以上であったため、本年度の成果となった。

Reason

ナノスケールから細胞スケール、メゾスケール、マクロスケールまで、当初の計画通りに研究成果が上がっている。さらに、当初の計画では最終年度に行う予定であった新規の治療法の提案についても、既に研究成果が出始めている。よって当初の計画以上に進展していると判断した。

Strategy for Future Research Activity

引き続き、ナノスケールからマクロスケールまでの各研究課題を推進する。細胞懸濁液に関する研究では、クラミドモナスを用いた実験を実施し、細胞スケールの走性の数理モデル化から、バイオリアクター内の流れの解明まで、複数の階層にまたがる研究課題を推進する。また、細胞懸濁液のスケールをまたがる流れを解析するための計算手法を調査し、我々の開発した手法のブラッシュアップも行う。さらに、得られた知見を応用し、細胞操作用のマイクロロボット等を開発する予定である。
