Outline of Annual Research Achievements

Rif1は、進化的に保存されており、酵母とヒトにおいて複製タイミングの制御に必須な役割をはたす。精製されたRif1はG4DNAに特異的に結合する。Rif1は、C端29aaに依存してG4DNAに特異的に結合するとともに、多量体を形成する。また同時に複数のDNAに結合することができる。分裂酵母ではRif1結合部位の機能はG4構造の形成に依存する。ChIPSeqで決定したマウスのRif1結合部位の中にもG4構造を形成する配列が存在する。これらの生化学的性質からRif1はゲノム上のG4DNAに結合しクロマチンファイバーを束ねてループ構造を形成しそれが複製や転写の機能ドメインの形成に貢献する可能性を提唱した。
Rif1ノックアウトマウスの解析から下記が明らかになった。
1.　B細胞の抗体産生特に免疫グロブリンのクラススイッチ（CSR）再構成に必要とされる。これはRif1がNHEJ（非相同末端結合）に必要であるという細胞レベルでの機能を反映するものである。2.　Rif1KOマウスではMbにおよぶ巨大なクラスター領域の一群の遺伝子の発現が著しく上昇した。同様なクラスター遺伝子発現の脱制御はRif1をノックダウンしたES細胞でも観察された。
またマウスES細胞におけるRif1の機能解析から下記が明らかとなった。
1.　Rif1はES細胞の自己複製には必須ではないが、全能性の維持には必要とされる。また効率のよいリプログラミングにも必要とされる。2.　Rif1のノックダウンにより2細胞期特異的遺伝子群の発現の増加が観察される。3.　Rif1は、ES細胞から３つの系列への分化の際の遺伝子発現制御に重要な役割を果たす。

Reason

ノックアウトマウスやノックダウン細胞の解析から高等動物でのRif1の機能が明らかになってきた。また、当研究室で開発した動物細胞発現精製システムにより全長2500aaにおよぶRif1タンパク質やそのtruncation formの精製が可能となり生化学的解析が大きく進んだ。
また分裂酵母から得られた知見を元に、動物細胞においてもRif1とG4 DNAとの関連を詳細に解析するという方向性が得られた。

Strategy for Future Research Activity

高等動物における　Rif1による複製や転写の制御と　Rif1結合部位との関連を明らかにする。
さらにこれらの知見をもとに、Rif1は複製や転写（そして組換えや修復も）を、クロマチン構造の改変を通じて、協調的に制御する可能性を検討する。
Rif1のさらに詳細な解析を通じて、その機能ドメインおよびリン酸化による制御の機構を解明する。
