海底下生命のセンサス：地球内部における生命活動の地理的空間分布と規定要因の解明

Outline of Annual Research Achievements

平成27年度は、世界各地の海洋底から掘削により採取された堆積物コア190試料から抽出された環境ゲノムDNAを対象に、マイクロ流体デバイス及びデジタルPCR法を用いて、バクテリアとアーキアの16S rRNA遺伝子断片の定量を行った。その結果、全堆積物コアに含まれるアーキア16S rRNA遺伝子量の全16S rRNA遺伝子量に対する割合は24.7%であり、2008年に研究代表者らが英科学誌ネイチャーに発表した従来手法による定量結果（35%～45%）を10%～15%程度下回る結果となった。そのうち、大陸沿岸の有機物に富む嫌気的な堆積物コア（144試料）に含まれるアーキア16S rRNA遺伝子の割合は平均29.4%であったのに対して、表層海水中の光合成基礎生産量が低い外洋から採取された堆積物コア（46試料）に含まれるアーキア16S rRNA遺伝子の割合は平均10.0%と優位に低い値を示した。さらに、同じ環境ゲノム試料からバクテリアとアーキアの16S rRNA遺伝子断片をPCR法により増幅し、次世代シーケンサーを用いて網羅的な塩基配列の解読を行った。本シーケンスを用いた微生物群集構造解析の結果、海底堆積物に存在する微生物生態系が、大陸沿岸域の嫌気的海底下生命圏と外洋の貧栄養海域の好気的海底下生命圏の2タイプに大別されることが判明した。これらの全球規模の海底下微生物群集の分析成果は、海底下生命圏における微生物群集の系統学的多様性やアーキアの存在量が、表層生命圏の光合成基礎生産等により供給される有機物の量や、現場環境の酸化還元状態に大きく支配されていることを示唆している。
一般的に、次世代シーケンサーを用いて標的核酸の網羅的なシーケンシングをする場合、PCR増幅効率等に起因するバイアスが生じるため、定量的な推定値を正確に得ることは難しい。その問題を解決するため、次世代シーケンサーを用いた標的遺伝子の網羅的な塩基配列の解読と定量を同時に行う新手法の開発に成功した。

Reason

全球的な海底下微生物の空間分布と遺伝学的特徴を解明するため、平成26年度には、世界各地の外洋及び大陸沿岸域から掘削により採取された凍結コア試料から環境ゲノムを抽出・精製した。平成27年度は、それらの試料を用いて、デジタルPCR法によるバクテリアとアーキアの16S rRNA遺伝子の定量と、次世代シーケンサーによる網羅的な塩基配列の解読を予定通りに終了した。さらに、次世代シーケンサーによる新規遺伝子解読・定量同時分析手法の開発にも成功した。従って、本研究課題はおおむね順調に進展していると思われる。

Strategy for Future Research Activity

平成28年度は、これまでに得られた16S rRNA遺伝子配列に基づく全球規模の微生物群集構造解析や統計学的マッピングを行うと同時に、海底堆積物に関わる様々な地質学的・物理学的・地球化学的データとの比較解析等を行い、海底下生命圏における微生物の伝播や消失、生命進化プロセス等に関する考察を行う。それらの研究成果を論文等にとりまとめ公表する。
