Outline of Annual Research Achievements

①Uncoupling Protein 1（UCP1）遺伝子のハプロタイプと非震え産熱（non-shivering thermogenesis : NST）の関連を調査した。生理実験をおこなった被験者（47名）のUCP1遺伝子座に存在する6つの一塩基多型（single nucleotide polymorphism: SNP）をタイピングした。その結果、ハプロタイプGnGTAnをホモ接合でもつ個体は、他のハプロタイプとのヘテロ接合やそれらをホモ接合でもつ個体と比べて、寒冷環境下で非震え産熱が有意に生じていることが示された。
②脂肪細胞の分化・機能に携わり、東アジア人の寒冷適応形質との関係が報告されているTRIB2遺伝子の一塩基多型について、日本人成人48名の脂肪組織試料での各種遺伝子転写への影響を調査した。その結果、適応型と考えられているアレルの保持者が、PPARGC1A等の脂肪組織での熱産生と関係した遺伝子群の転写量が高いことを見出した。また、モンゴル人96名を対象とした進化遺伝学解析から、3番染色体のCADM2-VGLL3遺伝子領域に過去の寒冷適応との関係を示唆する自然選択の痕跡を同定した。
③mtDNA多型と低圧環境下の生理的反応を明らかにするため、健常な男子大学生（D群18名、M7+G群10人）に対して実験を行った。室温28℃の低圧室を11.9Torr減圧し、9.7Torr（高度2500m相当）で15分維持、その後毎分9.7Torrで減圧して465Torr （4000m相当）を15分維持し、毎分11.1 Torrで与圧した。D群はM7+G群と比較して、末梢皮膚温が高く、気圧回復中に有意に高いSpO2を示した。すなわちmtDNAハプロタイプDは、急性低圧曝露中に末梢血流量を増やしSpO2を維持することが示唆された。
