| Outline of Annual Research Achievements |
本研究では、自然免疫-獲得免疫の連携に関わると考えられる化合物の探索と合成を基盤とした免疫制御機構の解明・展開を指向し、特に、種々の外因性・内因性分子の合成と機能解析、新規制御分子創製を中心に研究を進めている。本年度(H27年度）は前年度までの成果に基づき多くの興味深い結果を得ており、順次結果公表を進めている。まず、細菌由来の新規構造を持つ自然免疫受容体リガンドとして、免疫調節に関与し抗炎症作用に繋がると考えられる細菌表層成分の構造の詳細と活性に関する興味深い結果を得た。新規の天然型の外因性およひ内因性分子の化学合成法開発と機能解析については、真核生物に属する微生物の複合脂質の他、細菌の細胞壁成分の特異な修飾を持つ構造の合成と機能、あるいは細菌特有の脂質構造の新規合成を進め、世界で最初の全合成達成を含めた成果を得た。全合成を達成するため新規合成法開発については、光反応を利用したアミノアルコール合成法等、様々な天然物基幹構造に適用可能な汎用的な手法の開発を行った。合成化合物の免疫調節活性解析については，認識タンパク質-リガンドの直接的な相互作用評価系の確立あるいは導入を進め、免疫調節化合物の機能に関わる新しい知見を見出した。一方、感染症や癌等の疾患治療の基盤となる免疫機能調節可能な複合型分子については合成ワクチンのプロトタイプとなる分子の合成を行い、活性試験に供した。複合型活性構造に分子プローブとしての機能を付加した化合物を種々設計合成し、細胞内におけるイメージングにも成功している。当該年度において順調に成果が得られたことから、次年度はこられの知見に基づき、免疫制御機構解析と制御を可能とする分子の設計・合成および解析面での展開を進める。

当初の研究計画の目標を概ね順調に達成しており、一部、当初の計画以上に進展している部分もある。５件の科学雑誌への成果発表と５１件の学会発表の他、数件の論文投稿準備についても進めている。

| Strategy for Future Research Activity |
現状、研究計画にほぼ沿った形で順調に研究が推移しており、次年度は研究グループの人数も増えることから、新規化合物合成と解析法開発の両面でさらに強力に研究を推進する予定。
